ABSTRACT

The abnormal lipids metabolism is a critical pathological feature of coronary heart disease (CHD). Additional supplemental intake of polyunsaturated fatty acid (PUFA) has long been considered to be an effective strategy for preventing CHD, but more and more clinical trials have denied this view. Still, it is ambiguity for the specific mechanism of PUFA in CHD. The experimental programs are compliant with ethical principles for animal use and have been approved by the Animal Experiment Ethics Committee of Jinan University. In the present study, we established an animal model by intake of omega-6 PUFA combined acute myocardial ischemia to explore the mechanism of CHD. Intragastric administration of linoleic acid (LA) for 14 days, intraperitoneal injection of isoprenaline (ISO) was applied to induce acute myocardial ischemia for the animal model establishment. The animal ultrasound imaging system was used to detect cardiac function in vivo after ISO injection for 24 h. Serum and heart tissue samples were collected for the myocardial enzyme, phospholipidomics analysis and molecular biological detection. Compared to the LA group, the cardiac function showed that the left ventricular ejection fraction (EF%) and the left ventricular shortening fraction (FS%) decreased, aspaetate aminotransferase (AST), creatine kinase isoenzyme (CK-MB), and lactate dehydrogenase (LDH) increased in the LA + ISO mice. Compared to the ISO group, the phospholipidomics analysis showed that the PUFAs significantly were raised in the LA + ISO myocardium, and the content of oxidized phosphatidylethanolamine (ox-PE) changed most remarkable. Compared with the ISO group, the molecular biology detection showed that glutathione (GSH) and nicotinamide adenine dinucleotide phosphate (NADPH) were depleted, the end-products of ox-PE were increased, and the level of arachidonic acid 12/15-lipoxygenase (ALOX15) protein expression increased obviously. We suggest that ALOX15 mediated phospholipid peroxidation might be the critical mechanism of LA increased the susceptibility of myocardial ischemia injury. This study provides an experimental basis for whether PUFA could be used as an alternative treatment strategy for CHD prevention and provides a new intervention target for the early prevention strategy of CHD.

ABSTRACT

Using technical procedures, the fatty acid (FA) profile and solid fat content (SFC) of the Peruvian cultivar cacao beans CCN 51 and ICS 6 and the "optimal chocolate", obtained from the mixture of the first two, were determined to assess their quality. These cacao beans were found to have important nutritional values. The FA profile of the cacao beans were similar (p>0.05); however, in the FA profile, the 'optimal chocolate' had significant differences (p≤0.05) in terms of palmitic, arachidic and linolenic acid. The n6:n3 ratio for "optimal chocolate" was 12.0 ± 1.7. Cacao beans had the same SFC, and SFC was highly temperature dependent, as determined using a mathematical model for chocolate. The SFC of chocolate refers to hard cacao butter content at temperatures between 20 and 25°C, and solid fat was heat resistant from 25 to 30°C, which is considered valuable in trade chocolate production. The quality-related properties of these lipid fractions imparted nutritional and physical aspects to the optimal dark chocolate for human consumption.


La composición de ácidos grasos (CAG) y el contenido de sólidos grasos (CSG), de la fracción lipídica de los cultivares peruanos de cacao CCN 51 e ICS 6 así como del "chocolate óptimo", obtenido de las mezclas de las primeras dos, fueron determinados por técnicas analíticas para conocer su calidad. Estas variedades tuvieron valores nutricionales importantes. La CAG de los granos de cacao fueron similares, sin embargo la CAG del "Chocolate óptimo" tuvo diferencias significativas (p<0,05) para los acidos grasos palmitico, araquidico y linoleico. El ratio n6:n3 fue de 12,0 ± 1,7. El CSG de los granos de cacao fueron los mismos y tuvo una fuerte dependencia con la temperatura, también se definió un modelo matematico para el chocolate. El CSG le confiere al chocolate una consistencia dura a temperaturas de 20 a 25°C y resistentes al calor de 25 a 30°C, siendo tales propiedades una ventaja en la comercialización de chocolates. La calidad de estas fracciones lipidicas tuvieron aspectos nutricionales y fisicos en el chocolate oscuro para consumo humano.

ABSTRACT

The purpose of this study was to determine and compare fat composition and chemical properties of fish fillets of selected warm-water fish obtained from Straits of Malacca. A cold water fish, namely salmon was used for comparison. Moisture content, crude fat, fatty acids composition and chemical characteristics of fish fillets of Yellowstripe scad, Japanese threadfin bream and salmon were determined. Japanese threadfin bream fillet had highest moisture and crude fat contents, followed by fillets of Yellowstripe scad and salmon. A significantly strong and negative correlation was found between moisture and crude fat contents of these fish fillets. Fillets of Japanese threadfin bream and Yellowstripe scad also had higher total saturated fatty acids than total unsaturated fatty acids. Although salmon fillet had lowest percentage of saturated fatty acids, it had highest monounsaturated fatty acids and polyunsaturated fatty acids (PUFA) compared with the two warm-water fish. Palmitic acid and oleic acid were the major fatty acids in the fish fillets. Chemical properties of the oils extracted from the warm-water fish fillets were varied compared to salmon. The selected warm-water fish fillets offer favorable fatty acids composition and chemical properties, which can potentially be used as good sources of PUFA.


O objetivo deste estudo foi determinar e comparar a composição gordurosa e as propriedades químicas de filés de peixes selecionados de água quente obtidos no Estreito de Malaca. Um peixe de água fria, o salmão, foi usado para comparação. Foram determinados o teor de umidade, a gordura bruta, a composição de ácidos graxos e as características químicas dos filés de yellowstripe scad, sargo japonês e salmão. Os filés de sargo japonês apresentaram maior teor de umidade e de gordura bruta, seguidos por filés de yellowstripe scad e de salmão. Foi encontrada uma correlação significativamente forte e negativa entre a umidade e o teor de gordura bruta desses filés de peixe. Filés de sargo japonês e de yellowstripe scad também apresentaram ácidos graxos saturados totais mais altos do que os ácidos graxos insaturados totais. Embora o filé de salmão tenha menor porcentagem de ácidos graxos saturados, ele apresentou os maiores ácidos graxos monoinsaturados e ácidos graxos poliinsaturados (PUFA em comparação com os dois peixes de água quente. O ácido palmítico e o ácido oleico foram os principais ácidos graxos dos filés de peixe. As propriedades químicas dos óleos extraídos dos filés de peixe de água quente foram variadas em comparação ao salmão. Os filés de peixe de água quente selecionados oferecem composição favorável de ácidos graxos e propriedades químicas, que podem ser potencialmente usadas como boas fontes de PUFAs.

ABSTRACT

Objective To investigate the effects of enteral immunonutrition supplemented with omega-3 polyunsaturated fatty acid (ω-3 PUFA) on the incidences and severity of ventilator associated pneumonia com- plications, inflammatory response, and the prognosis in patients with severe traumatic brain injury (sTBI) un-dergoing ventilator therapy. Methods From January 2015 to June 2017, 64 patients of sTBI were selected and randomly divided into experimental group (ω-3 PUFA, n=32) and control group (n=32). White blood cells (WBC) and the proportion of neutrophils (N%) were evaluated by cell analyzer. Meanwhile, the serum levels of C-reactive protein (CRP) and procalcitonin (PCT) were tested with enzyme linked immunosorbent assay. After 14-days treatment, the Glasgow coma scale (GCS) score, APACHE Ⅱ score, clinical pulmonary infec-tion score (CPIS), pulmonary function and prognoses were also compared between the two groups. Results As compared with the control group, the experimental group had lower incidences of ventilator associated pneumonia (66% vs. 56%, P=0. 048), reduced degree of lung infection and lower CPIS (8. 25±0. 60 vs. 7. 47±0. 53); higher lung function indexes [FVC: (2. 89±0. 19) L vs. (3. 46±0. 22) L, P=0. 010;FEV1: (2. 35±0. 16) L vs. (2. 84±0. 24) L, P=0. 040; FEV1/FVC %: (49. 11±3. 34)% vs. (56. 00± 2. 64)%, P=0. 038) ]; lower serum levels of inflammatory responses [WBC: (11. 83±0. 74) ×109/L vs. (9. 51±0. 90) ×109/L, P=0. 029; N%: (79. 11±1. 51)% vs. (72. 71±1. 16)%, P=0. 041; CRP:(85. 15±8. 42) mg/L vs. (63. 96±5. 72) mg/L, P=0. 001; PCT: (6. 43±0. 47) μg/L vs. (4. 83±0. 39) μg/L, P=0. 013] 14 days after enteral immunonutrition supplemented with ω-3 PUFA. As compared with the control group, the experimental group received better prognosis with GCS scores increasing ( 8. 69 ± 0. 41 vs. 9. 52±0. 59, P=0. 038), APACHE Ⅱ scores decreasing (14. 74±1. 01 vs. 12. 68±0. 89, P=0. 049), the time of mechanical ventilation [ (13. 23±1. 17) d vs. (10. 88±1. 24) d, P=0. 024] and the hospitalization days [ (23. 29±2. 45) d vs. (18. 42±1. 96) d, P=0. 012] reduced on the 14th day, mechanical ventilation withdraw rate within 14 days increasing [24/32 (75%) vs. 27/32 (84%), P=0. 030] on the 14th day. Conclusion Enteral immunonutrition supplemented with ω-3 PUFA can effectively reduce the incidence of ventilator associated pneumonia, alleviate the degree of infection and the inflammatory response in patients with sTBI undergoing ventilator therapy possibly improving condition and prognosis, which is worthy of being widely used.

ABSTRACT

ABSTRACT: This study aimed to present a literature review about the characteristics, applications, and potential of avocado (Persea americana). Avocado is considered one of the main tropical fruits, as it contains fat-soluble vitamins which are less common in other fruits, besides high levels of protein, potassium and unsaturated fatty acids. Avocado pulp contains variable oil content, and is widely used in the pharmaceutical and cosmetics industry, and in the production of commercial oils similar to olive oil. This fruit has been recognized for its health benefits, especially due to the compounds present in the lipidic fraction, such as omega fatty acids, phytosterols, tocopherols and squalene. Studies have shown the benefits of avocado associated to a balanced diet, especially in reducing cholesterol and preventing cardiovascular diseases. The processed avocado pulp is an alternative to utilize fruits, which can be used in various value-added food products. Fluid extract of the avocado leaves is widely used in pharmaceutical products, mainly due to the diuretic characteristic of the present compounds in plant leaves. With the increasing research supporting the nutritional characteristics and benefits of avocado, the tendency is to increase the production and exploitation of this raw material in Brazil, as also observed in other countries.


RESUMO: Objetivou-se com este trabalho apresentar uma revisão da literatura sobre as características, aplicações e potencialidades do abacate (Persea americana). O abacate é considerado um dos principais frutos tropicais, pois possui as vitaminas lipossolúveis que, em geral, são deficientes nas outras frutas. Além destas, contém proteínas e elevados teores de potássio e ácidos graxos insaturados. Apresenta quantidade variável de óleo na polpa, sendo esta vastamente utilizada nas indústrias farmacêutica e de cosméticos, e na obtenção de óleos comerciais similares ao azeite de oliva. Esse fruto tem sido reconhecido por seus benefícios à saúde, especialmente em função dos compostos presentes na fração lipídica, como ácidos graxos ômega, fitoesterois, tocoferois e esqualeno. Estudos têm demonstrado os benefícios do abacate associado a uma dieta balanceada, principalmente, na redução do colesterol e na prevenção de doenças cardiovasculares. A polpa do abacate na forma processada é uma alternativa para aproveitar os frutos, passível de ser utilizada em diversos produtos alimentícios de maior valor agregado. O extrato fluido das folhas do abacateiro é muito utilizado em produtos farmacêuticos, principalmente, pelo caráter diurético de compostos presentes nas folhas da planta. Com o aumento das pesquisas que comprovam as características nutricionais e os benefícios do abacate, a tendência é aumentar a produção e exploração dessa matéria-prima no Brasil, como já acontece em outros países.

Introduction. Monos Group, Drug Research Institute is starting to investigate of Ellipin preparationfrom the mid-1990s, Ellipin has anti cancer activity in liver and several studies were investigated withscientists from Japan and China. Especially Hayashi K., Khurelbaatar L and Ambaga M were determinedanti-cancer action of the preparation and they were explained of mechanism of action, which apoptosisis seduced by influence of unsaturated fatty acids in tumor cells. However, changes of fatty acidscomposition at production stage were did not study yet. Therefore, we studied that composition of fattyacids in different term of production stage and compared of Ellipin dense substance.Materials and Methods. Samples of study were collected from production stage of “Ellipin” series130304, which was tacked in 48th hour, 120th hour of production. Each sample was dried at freezedryer “Labconco freezone12L” in Drug Research Institute. Total lipids of sample were extracted withchloroform: methanol (2: 1 v/v) according to Folch et al. Fatty acid methyl esters were analyzed usingAgilent Packard Gas Chromatograph (GC) (Model HP-6890 Agilent Packard) with mass-spectrumdetector (Model HP MSD 5973N) of Buryat State University, in Ulan-Ude.Results. Ellipin preparation is derived from bovine liver, and which is based on homogenization of bovineliver for isotonic. In this process, unsaturated fatty acids were extracted in organic solution. We studiedchanges which saturated and unsaturated fatty acids of bovine liver in process of homogenization andconsist of each fatty acid contents of end product. Results have shown that unsaturated fatty acidswere decreased by 0.4-44% till 120th hour of homogenization process. While, there were decreasedby 4-12% in the end product, although, ω-6 fatty acids were increased by 13.1-38.4%. Moreover, 25saturated fatty acids and 12 unsaturated fatty acids were detected in the Ellipin dense substance (endproduct). Hence, 67.5% of total fatty acid was saturated fatty acids, 32.5% was unsaturated fatty acidsin the Ellipin dense substance. Resent results and results of previous studies indicated that Ellipindense substance may contains saturated fatty acids on in average 50.34%, unsaturated fatty acids onin average 49,32%, respectively.Conclusion. Proportion of saturated and unsaturated fatty acids in Ellipin production was about 2:1.Saturated fatty acids and unsaturated fatty acids were found 25 and 12, respectively. Saturated fattyacids were gradually decreased and unsaturated fatty acids were slowly increased in production period,which from 48th hour of production-conveyer till end product. Moreover, content of ω-3-6-9 fatty acidswas consist 83,9-87,5% of total unsaturated fatty acid.

Background Researches determined that ELOVL4 gene is a disease-causing gene of Stargard tmacular dystrophy and is a elongation enzyme of very long chain fatty acids.Stargardt type Ⅲ(STGD3) may be associated with the metabolism of extensive enzyme of very long chain fatty acids.To explore the effect of ELOVL4 in STGD3 and its relationship with the metabolism of extensive enzyme of very long chain fatty acids is of important clinical significance.Objective This study was to determine the role of ELOVL4 gene for the pathogenesis of STGD3.Methods Recombinant adenovirus type 5 carrying mouse ELOVL4 gene and green fluorescent protein (GFP) was transfected into pheochromocytoma cells (PC12 cells),and then the cells were divided into PC12 group,PC12+Ad5-GFP group and PC12+AdS-ELOVL4 group.Ad-GFP or Ad-ELOVL4 was added into the culture medium for 24 hours with the virus plasmid 1 × 104-2× 104 pfu/ml.Expression of ELOVL4 mRNA in the PC12 was quantified by quantitative real time PCR(qRT-PCR) and was described as relative value to the expression of RPL19.ELOVL4 protein was assayed by Western blot.The transfected cells were treated with arachidonic acid (AA,20:4n6),eicosapentaenoic acid (EPA,20:5n3) and docosahexaenoic acid (DHA,22:6n3) individually for 48 hours.The cells were collected,and total lipids were extracted,and fatty acid methyl esters were prepared and analyzed by gas chromatography-mass spectrometry (GC-MS).Results The relative expression levels of ELOVL4 mRNA in PC12 cells in the PC12+Ad5-ELOVL4 group,PC12+Ad5-GFP group and PC12 group were 0.833± O.138,0.027t±0.002 and 0.024 ±0.002,with a significant difference among the 3 groups (F =102.700,P =0.000),and relative expression levels of PC12+Ad-ELOVL4 were 30-40 folds more than those in the PC12 group and the PC12+Ad-GFP group.Western blot assay showed a stronger response band for ELOVL4 protein in the PC12+Ad-ELOVL4 group.GC-MS found that abundant polyunsaturated fatty acids (C28-C38) were synthesized by PC12 cells in the PC12+Ad-ELOVL4 group,with the more levels in C34 and C36.Conclusions ELOVL4 can promote the synthesis of C28-C38 polyunsatured fatty acid in PC12 cells,which offers a novel clue for the treatment of STGD3.

ABSTRACT

Objective To explore the therapeutic effect of immune regulating nutrition on severe acute pancreatitis (SAP). Methods The clinical data of 80 patients with SAP who were admitted to the Union Hospital of Huazhong University of Science and Technology from February 2008 to October 2009 were prospectively analyzed. The 80 patients were equally divided into conventional nutrition (CN) group and immune regulating nutrition (IRN) group. Patients in the CN group received CN support, while patients in the IRN group were supplemented with ω-3 polyunsaturated fatty acid (ω-3 PUFA ) and glutamine at the basis of CN support.APACHE Ⅱ scores were evaluated at day 1, 4, 7, 10, 14 in the course of treatment. The differences in the incidence of pulmonary and abdominal infection, sepsis, and other clinical parameters including operation intervention, mortality, length of ICU and hospital stay between the two groups were compared. All data were analyzed using the t test and chi-square test. Results The APACHE Ⅱ scores of patients in the IRN group were 15.3 ± 1.8 and 9.0 ± 1.8 at day 4 and 7, which were significantly lower than 20.0 ± 2.7 and 13.3 ± 2.4 in the CN group, respectively (t = 3.3, 2. 8, P < 0.05). The APACHE Ⅱ scores of patients in the IRN group at day 10and 14 were also lower than those in the CN group, while there was no significant difference (t =0. 7, 0. 6, P>0.05). The incidences of pulmonary and abdominal infection and surgical intervention rate in the IRN group were 18% (7/40), 13% (5/40) and 5% (2/40), which were significantly lower than 38% (15/40), 33% (13/40)and 20% (8/40) in the CN group, respectively (χ2 =4.0, 4.6, 4.1, P<0.05). The incidence of sepsis in the IRN group was 5% (2/40), which was lower than 8% (3/40) of the CN group, while there was no significant difference between the two groups ( χ2 =0.0, P >0.05 ). The average lengths of ICU and hospital stay in the IRN group were (5.4 ± 1.6 ) days and ( 38.6 ± 9.3 ) days, which were significantly lower than ( 7.8 ± 2.8 ) days and (43.1 ± 1 1.6) days in the CN group, respectively ( t = 2. 7, 3. 7, P < 0.05 ). The morality was 5% (2/40) in the IRN group and 8% (3/40) in the CN group, with no significant difference between the two groups (χ2 =0.0,P >0.05). Conclusion CN support supplemented with ω-3 PUFA and glutamine can adjust the inflammation reaction of SAP and sustain the normal immune function, which is helpful in decreasing the incidence of severe infection.

ABSTRACT

Objective To study the effect of unsaturated fatty acid of Pinus koraiensis on lipid metabolism in rats. Method Unsaturated fatty acid was extracted and concentrated from Pinus koraiensis by supercritical fluid extraction and urea inclusion method. 40 rats were divided into 8 groups randomly. Except normal control group, the other groups were fed high fat diet. Normal control and obesity control groups were fed i.g. normal saline,Seed oil control group was fed i.g. pine nut oil and the other groups were fed i.g. 0.4, 0.8, 1.2, 1.6 and 2.0 g/kg bw unsaturated fatty acid. Body weights were weighed every week. After 4 w, the rats were killed, the serum total triglycerides(TG), total cholesterol(TC), high density lipoprotein cholesterol (HDL-C) and hepatic lipase(HL), lipoprotein lipase(LPL), liver lipofuscin (LP) were assayed. Results Compared with obesity control group, the body weight, body fat ratio, serum TG, LP and TC level were reduced remarkably. HL and total lipase (LA) level enhanced remarkably. Conclusion The unsaturated fatty acid of Pinus koraiensis could enhance the lipid metabolism in obese rats.

0.999).The minimumdetectable limits of ALA,LA and DHA were all 0.2 ?g/ml,and AA was 0.4 ?g/ml.The average recovery of the method was 92.65%~100.70% and the RSD was lower than 5.67%.Conclusion:The HPLC method in this assay is rapid,direct and sensitive,which can be successfully applied to quantity the detection of polyunsaturated fatty acid of different biological samples as well as in clinic.

ABSTRACT

Aim To study cytotoxic effects and mechanisms of unsaturated fatty acid from mao er yan of gansu origin on BEL-7402 strain cells.Methods The essential components of unsaturated fatty acid from mao er yan of gansu origin were determined by gas chromatography-mass spectrometry and cytotoxic effects of un-saturated fatty acid on BEL-7402 strain cells were determined by using flow cytometry,methods of MTT [3-(4,5-dimethyl thiazol-2yl)-2,5-diphenyltetrazolium bromide] and electron microscope scanning after 48 hours of the unsaturated fatty acid from mao er yan of gansu origin treatment at concentrations of 0.2,0.8 and 3.2 mg?L-1,respectively.Results The unsaturated fatty acid from maoer yan of gansu origin had significantly cytotoxic effects on BEL-7402 strain cells and inhibitory rates for proliferation of BEL-7402 strain cells were 34.6%、44.8% and 70.7% at those three concentrations,respectively.50% inhibiting concentration(IC_ 50)was 0.812 mg?L-1.Apoptosis rates were 18.6%,22.7% and 24.2%,respectively,and occurred cell membrane microvilli breakdown and cell rupture,compared to those of the control group.Conclusion The unsaturated fatty acid from mao er yan of gansu origin has significantly cytotoxic effects on BEL-7402 strain cells and their mechanisms may be related to inhibit cell division,destroy the structure of cell membrane and mitochondria.

ABSTRACT

Objective To screen antarctic bacteria producing EPA and analyse its composition of fatty acids.Methods Two hundred strains of bacteria were isolated from ice and soil of the Antarctic.The total lipid was extracted by using a modified one-phase Bligh and Dyer extraction,and screened by means of GC using fish oil as standard.In the end,eight strains of bacteria were screened which had high content of EPA or DHA.Among them,one bacterium(NJ136) which had the highest content of EPA was obtained,its total lipid was extracted by means of Soxhlet extractor,and the compositions of fatty acids were analysed by means of GC-MS.Results and conclusion The relative contents of EPA and total unsaturated fatty acids of NJ136 were 6.66% and(51.92%) respectively,the content of total fat,EPA and unsaturated fatty acids of dry bacteria weight were 7.94%,5.3 mg?g~(-1) and 41.2 mg?g~(-1),respectively.

ABSTRACT

The transport of α-methyl-D-glucoside and two aminoacids, L-phenylalanine and L-leucine by a temperature sensitive fatty acid requiring mutant of Salmonella typhimurium was studied under conditions of supplementation with cis or trans-unsaturated fatty acids. The results of such experiments definitely establish a relationship between the fatty acids composition of the membrane and the transport property of the cells. Cells grown in the presence of trans–unsaturated fatty acids cannot transport so efficiently as compared to the cis-unsaturated fatty acid-grown cells except linolelaidic acid, a trans-transunsaturated fatty acid. Protein: phospholipid ratio of the membrane also varies significantly under such conditions. The affinity of L-phenylalanine transport carrier for the substrate changes remarkably in cells grown in the presence of different cis or trans-unsaturated fatty acids and indicate the possible role of membrane lipids in membrane assembly as well as regulation of the activity of L-phenylalanine transport system.
